Overview

R)-1-[3,5-bis(trifluoromethyl)phenyl]ethanol is an important chiral intermediate for the synthesis of the antiemetic drug Aprepitant

Uses

(R)-1-[3,5-bis(trifluoromethyl)phenyl]ethanol is mainly used to treat depression and chemotherapy-induced vomiting; can prevent acute and delayed nausea and vomiting caused by the first and subsequent repeated chemotherapy; API aprepitant/aprepitant intermediate; aprepitant morpholine hydrochloride side chain.

Preparation

Dissolve 3,5-bis(trifluoromethyl)acetophenone in methanol, add 2% sodium borohydride in sodium hydroxide solution dropwise under ice bath, react for 2 hours, adjust with hydrochloric acid until no bubbles emerge, distill to remove methanol solvent, cool, extract with dichloromethane, dry and concentrate to obtain racemic product.
