What makes it different on the bench

Crospovidone, registered under CAS 25249-54-1, is one of those materials that rarely makes the headlines yet shows up in almost every pharmacy shelf. Chemically it is cross-linked polyvinylpyrrolidone, usually written as crospovidone or PVPP. Unlike the linear PVP you meet in solutions and coatings, this version is built into a three-dimensional network through cross-linking, and that single change explains the behaviour formulators actually care about.

The first thing you notice is that crospovidone does not dissolve in water. Drop it into a beaker and it just sits there, swelling instead of going into solution. That swelling is fast and volume-driven. When a compressed tablet takes in water, the crospovidone particles absorb liquid, expand, and push the surrounding mass apart. The tablet breaks along weak points and the granules fall apart within seconds. For oral solid dosage this is the job of a superdisintegrant, and crospovidone is one of the few names most formulators reach for first.

A useful property is that it does not gel. Some disintegrants form a sticky layer that slows everything down; crospovidone stays porous and keeps wicking liquid inward. That is why it works well even with drugs that are themselves poorly soluble, where you do not want the disintegrant to become part of the dissolution bottleneck.

Typical specifications buyers ask for

In practice the numbers checked at incoming QC are fairly consistent across suppliers. Bulk density usually lands between 0.2 and 0.4 g/cm³ for the free-flowing grades. The pH of a 10% aqueous dispersion sits around 5 to 8. Nitrogen content by Kjeldahl is commonly reported in the 11.0 to 12.8% range, which is a quick check that the material is what the certificate claims. Residual vinylpyrrolidone monomer is kept low, typically under 10 ppm for pharma grades that follow USP/NF or Ph. Eur. Particle size is the lever formulators pull most often: finer grades give faster disintegration but can affect powder flow and tablet hardness, while coarser grades are gentler on compaction.

Where it actually gets used

Tablets are the obvious case, but the material has a second life in beverages. In winemaking and brewing, crospovidone is used as a fining agent. It binds polyphenols and tannins, the compounds that cause haze and astringency as the product ages. A measured dose, often in the tens of grams per hectolitre, clears the liquid and improves stability. The same binding behaviour is why it turns up in some cosmetic masks and cleansers, where it helps with oil control.

For tablets, typical addition levels run from 2% to 5% by weight. It performs in both direct compression and wet granulation, and it can be added intra-granular or extra-granular depending on the release profile you want. Because it is inert and has almost no taste or odour, it rarely interferes with the active ingredient or the patient experience.

Handling and storage without surprises

Crospovidone is hygroscopic, so the main rule is to keep the container closed and the storage area dry. Once it picks up moisture the flow properties drop and you get bridging in the feed frame. Stored at room temperature in a sealed drum it keeps well past two years. Like any fine powder, avoid creating dust: use local extraction or a downdraft table when handling open bags, and follow the standard PPE for nuisance dust.

Sourcing and what to ask a supplier

When you request a quote, the documents matter as much as the price. Ask for a current certificate of analysis tied to the batch, an MSDS, and a technical data sheet that states the pharmacopoeia grade. If your market requires it, confirm food or beverage grade separately from pharma grade, since the specifications are not identical. Particle size distribution and loss on drying are the two values most likely to vary between lots, so pin them down in the specification agreement rather than assuming they match the catalogue.

For teams scaling a formulation, consistency batch to batch is the real cost driver. A supplier that holds particle size and monomer residue steady will save more in validation time than a small discount on the first shipment ever will. That is the part worth checking before you commit.
